In order to ensure that the results obtained by the blood cell analyzer can reflect the real situation of patients as far as possible, the following aspects must be paid attention to when using the blood cell analyzer: 

1. Blood sample: because the external factors have little influence on the venous blood, the composition is relatively stable, the accuracy of the test results is high and repeatable, therefore, except for infants, the proposed blood should be taken by venous blood. If the collection of no tip blood, pay attention not to local over-extrusion, avoid blood mixed with a lot of tissue fluid, and easy to activate the coagulation system to produce local coagulation, resulting in errors in the detection results; The first drop of blood should be discarded because of the instability of the cellular component, and the second drop of blood should be tested.

2. Anticoagulation: the use of citrate anticoagulant too long, easy to crystallize, cell morphology changes, affect the accuracy of counting results; oxalate easy to platelet aggregation, and can change the morphology of white blood cells, excessive heparin can cause leukocytes agglutination and thrombocytopenia, EDTA-2Na is less soluble than EDTA-2K, and platelet agglutination is more likely. Therefore, the International Committee for Haematological Standards (ICSH) published a document in 1993 recommending the use of EDTA-2K as an anticoagulant for hematology analyzers in doses of 1.5-2.2 mg/ml of blood. 

3. The blood was sealed with a plug and kept at room temperature for no more than 6 hours.

4. Dilution: the diluent and the sampling tube should be calibrated. After sucking blood, the blood outside the suction tube should be completely wiped clean. Hemodilution should be determined as soon as possible, otherwise easy to cause“Dilution hemolysis.”. 

5. Mixing: it is important to mix well before testing. If there is no rotary blender, mix it upside down at least 8 times. 6. Reagents: the hematology analyzer requires very strict reagents, requiring a strict osmotic pressure standard, stable conductivity, high standards of purity and no corrosion of the instrument pipeline and valve. Therefore, the best choice of hemolytic, diluent and cleaning agents, such as the original factory supporting products.

Seven. Classification of white blood cells: first of all, it must be clear that up to now, no matter how advanced blood cell analyzer in the world, the classification of white blood cells is only a screening method, and can not completely replace manual microscopy classification. We must resolutely correct the mistaken idea that some units lose microscopic examination when they use blood cell analyzer. 8.
